Cover Cover: Gene delivery of low-molecular-weight caldesmon (l-CaD) protects cytoskeletal cell membrane integrity against adenoviral infection independently of myosin ATPase and actin assembly. Human l-CaD was expressed in avian fibroblast DF-1 cells, a spontaneously immortalized but nontransformed cell line null for l-CaD, using calcium phosphate coprecipitation at a multiplicity of infection of 200. Colocalization of human l-CaD with avian microfilaments was determined using immunofluorescence imaging. Cells were coimmunostained with a mouse IgG anti-l-CaD primary antibody, an Oregon Green-conjugated goat anti-mouse secondary antibody, and Texas red phalloidin to visualize l-CaD and microfilaments, respectively. Images were obtained using FITC and rhodamine filters at ×100 magnification. Images depicting l-CaDstained cells (green) and actin-stained cells (magenta) from the same field of view were then overlaid as shown to display colocalization (white- or lighter magenta-colored filaments).
