Fig.5. Cellular responses to shear are dependent on phospholipase
C (PLC)-activated, IP3-mediated intracellular
Ca2+ release. PLC inhibitors neomycin (10 mM) or U-73122
(10 µM) were added to MC3T3-E1 cells 30 min before application of 12 dyn/cm2 shear for 1 h. A: illustrates
typical response of stress fiber formation and c-fos expression in
untreated cells to shear. PLC inhibition with neomycin (B)
or U-73122 (C) completely inhibits these responses. Response
of MC3T3-E1 cells pretreated with inactive isoform of U-73122, U-73343
(D; 10 µM), was not significantly different from control
cells. COX-2 production was affected in a similar manner.
